摘  要:桶装纯净水中存在一定量的微生物,对3个不同品牌桶装纯净水中的菌落总数按照国家标准进行测定,其菌落总数分别为9.8、16.8cfu/mL和33.4cfu/mL。采用水琼脂培养基和10%浓度的牛肉膏蛋白胨培养基,从中还得到2株细菌,分离纯化后,用牛肉膏蛋白胨培养基扩大培养,通过PCR扩增出其16SrDNA基因,经测序比对后进行聚类分析,确定2株细菌分别属于柄杆菌属(Caulobacter)和罗尔斯通氏菌属(Ralstonia)。There are many microbes in the barreled pure water. Total number of colony of barreled pure water were done accord to national standard. The colony count of the three different sample were 9.8, 16.8 cfu/mL and 33.4 cfu/mL respectively. Two bacteria were isolated from the barreled pure water by using of 10% beef extract-peptone medium and water-agar medium. Both of them were purified by the beef extract-peptone medium.16S rDNA genes of the two bacteria were copied by PCR and then sequenced by Sanger method. After blasted on NCBI and cluster analysed by MEGA, the two bacteria were defined belong to Caulobacter and Ralstonia.
