摘  要:目的:研究贻贝蒸煮液中多糖的免疫学活性及对淋巴瘤细胞Raji的生长抑制作用。方法:通过乙醇沉淀、Sevage法除蛋白、活性炭脱色,再经琼脂糖凝胶Sepharose 4B柱层析和透析,分离纯化得到贻贝蒸煮液多糖MJPs-1和MJPs-2,并对其免疫学活性、体外抗肿瘤活性以及细胞毒性进行测定。结果:MJPs-1对小鼠T、B淋巴细胞有显著的正向调节作用;低剂量的MJPs-2对小鼠B淋巴细胞有显著的负向调节作用,而高剂量的MJPs-2对小鼠T淋巴细胞表现出正向调节作用。此外,MJPs-1和MJPs-2均能显著抑制Raji细胞的增长且对细胞毒性较小。结论:贻贝蒸煮液多糖MJPs-1和MJPs-2具有免疫调节作用和抑制Raji细胞增殖的活性,贻贝蒸煮液多糖的低毒性使其在保健品的开发和结合其它强抗癌制剂以降低药物的细胞毒性方面具有极大的应用价值。Objective:To study the extraction of Mussel Juice Polysaccharides (MJPs), their immunological activities and anti-tumor promoting activities. Methods:The MJPs was extracted by alcohol fractional precipitation, free proteins and colors were removed by Sevage method and charcoal, respectively. MJPs-1 and MJPs-2 are isolated and purified by Sepharose 4B column and dialysis membrane. The immunological activities, anti-tumor promoting activities and cytotoxicity of purified MJPs-1 and MJPs-2 are tested. Results:MJPs-1 showed significant positive adjusting effect on T lymph cells and B lymph cells of mice. MJPs-2 exhibited significant negative adjusting effect on B cells in low concentration and adjusting effect on T lymph cells in high concentration. Both of them showed the inhibitory effects on EBV activation and low toxicity on Raji cells. Conelusion:MJPs-1 and MJPs-2 display immunological activities and anti-tumor promoting activities with low toxicity on Raji cells, which indicated that the Mussel Juice Polysaccharides may be valuable on developing the functional foods and reducing the cytotoxicity when added in other strong anti-cancer agents with high cytotoxicity.
