模拟酸雨对噬藻体PP失活率、野生藻的吸附率、裂解周期及释放量的影响

摘  要:研究不同pH值的模拟酸雨对噬藻体PP失活率及噬藻体PP感染野生藻的吸附率、裂解周期及释放量的影响。用离心法测定了噬藻体PP对宿主藻的失活率、吸附率,用一步生长曲线法获得了噬藻体PP对宿主藻的裂解周期及释放量。结果表明,在pH4.0~7.0的条件下:噬藻体PP7h的失活率分别为81.9%、74.8%、72.8%、67.1%,且pH4.0组的失活率显著高于与pH7.0组(对照组);噬藻体PP对野生藻的吸附率在45min时达到最大值,分别为54%、56.9%、67.9%、68.8%,各处理组的最大吸附率均显著低于对照组;噬藻体PP感染野生藻的潜伏期均介于30~90min之间,释放量分别约为42、52、29、104PFU·mL-1,各处理组与对照组的释放量存在极显著差异。表明模拟酸雨对噬藻体感染动力学过程的影响具有重要的意义。The decay rate, adsorption rate, lysing cycle and burst size of cyanophage PP infecting the wild Filamentous cyanobacteria was studied in the lab condition . Acid solution was prepared with H2SO4 and HNO3 adjusting to pH4.0, pH5.0 , pH6.0 and pH7.0 respectively for control . The rate of inactivation and the absorption was measured by using the centrifugal method. Lysing cycle and burst size were obtained with one step growth curve method. In the simulated acid rain condition, the results indicated that the decay rates were 81.9%、74.8%、72.8%、67.1% respectively, and the significant level of decay rates were found in pH4.0 treatment comparing with the control. At time ponit of 45 min the adsorption rates reached the maxium, they were 54%、56.9%、67.9%、68.8% respectively . The adsorption rates of treated groups were significantly lower comparing with the control. The latent period was about 30min to 90min and the burst sizes were 42、52、29、104 PFU per cell respectively. The burst sizes of treated groups were significantly different from the control. These results indicated that simulated acid rain plays a very importsnt role in the decay rate and the infection kinetics of cyanophage PP.
