食管癌组织中Cyr61的表达及临床意义

摘　　要：目的探讨人食管癌组织中Cyr61 mRNA、蛋白表达及其与临床参数的关系。方法采用实时荧光定量PCR和免疫组化染色法对186例食管癌患者的食管癌组织和癌旁正常食管组织中Cyr61 mRNA蛋白表达水平进行测定。结果食管癌组织中Cyr61 mRNA、蛋白表达水平均显著高于配对的癌旁正常食管组织(P均= 0.000)。Cyr61 mRNA表达水平与食管癌患者淋巴结转移、浸润深度、肿瘤大小、病理分级有关(P<0.01),Cyr61蛋白表达水平与患者吸烟及肿瘤淋巴转移、家族史、浸润深度、肿瘤大小和病理分级有关(P<0.05)。多因素Logistic回归分析显示,淋巴结转移、肿瘤大小、浸润深度均是Cyr61 mRNA和蛋白表达的危险因素(P<0.05)。结论Cyr61 mRNA在食管癌发展过程中起重要作用,其表达的蛋白产物可能作为临床评估治疗的重要靶蛋白。Objective To investigate Cyr61 mRNA and protein expression levels and their relations to clinical and pathological features in human esophagus carcinoma. Methods To be quantified Cyr61 expression levels in 186 case esophageal cancer issues and their matched normal esophagus tissues by using real-time PCR assay and immunohistochemistry staining. Results The Cyr61 mRNA and protein expression levels of esophageal cancer tissues were significantly higher than that in the matched normal esophagus（ both P =0.000）. The Cyr61 mRNA expression levels had significant association with metastasis, infiltrate depth, tumor size and tumor stage （P 〈 0. 01 ）. Cyr61 protein expression levels also had significant association with smoke, metastasis, family history, infiltrate depth, tumor size and tumor stage（P 〈 0.05）. Multiple factor Logistic regression showed that metastasis,tumor size,infiltrate depth were all risk factors（P 〈 0.05）. Conclusions Cyr61 may play a role in the progression of esophageal cancer, and their proteins might serve as valuable targets for early diagnosis and therapeutic intervention.
