摘 要:目的 应用肿瘤坏死因子 (TNF)受体 基因敲除突变型鼠 ,探讨 TNFα介导郎格罕细胞(L C)向局部淋巴结迁移、引发免疫应答过程中 ,其两种膜受体 :TN F受体 和 TN F受体 所起的信号转导作用。方法 以免疫荧光标记及流式细胞分析技术分别观察 TNF 受体 缺失鼠和经抗 TNFα中和性抗体预处理以阻断其体内 TNFα生物学效应的 TN F受体 缺失鼠在 FITC接触致敏后 ,L C自皮肤向淋巴结的迁移情况 ,并与野生型鼠对照。结果 经致敏后 ,TNF受体 缺失鼠淋巴结内出现的 FITC携带细胞(证实为 L C)数量与野生型鼠无明显差异 ;而经抗 TN Fα抗体预处理的 TN F受体 缺失鼠引流淋巴结内L C数量明显少于未经 TNFα抗体预处理的对照组。结论 TN F受体 在介导 L C迁移中并非必需 ;在TNF受体 缺失情况下 ,TN F受体 的信号转导对 L C的迁移可能发挥关键作用。Objective To elucidate the role of signal transduction TNF receptor I(TNF R1) and TNF receptor (TNF R2 ) in L angerhans cells (L C) m igration in im muno-responses,TNF R1gene-knock- out mutant m ice were used in this study.Methods The L C m igration from skin to regional lymph nodes after F ITC sensitizatioon in both the TNF R1-deficient m ice and the TNF R1-deficient m ice pretreated with neutralizing antibody to TNFα w ere exam ined by im munolabelling and F CM,and TNF R1+ / + m ice were used as control. Results Following F ITC sensitization there was no significant difference between the num ber of F ITC+ cells(L C) found in lym ph nodes from TNF R1-deficient mice,and from the wild type (TNFR1+ / + ) m ice,and the number of L C found in the draining lymph nodes from anti-TNFα antibody pretreatm ent TNF R1-deficient mice was significantly lower than that of the m ice not pretreated by anti- TNFα antibody. Conclusion It suggests that TN FR1is not necessary in m ediating L C m igration and TN - F R2 might be crucial in signal transduction in TNF R1-deficient m ice.
