β-胡萝卜素和亚硒酸钠体外抗肿瘤作用的实验研究

摘　　要：方法：采用细胞株体外培养技术，研究β－胡萝卜素和亚硒酸钠（Ｎａ２ＳｅＯ３）单独／联合对人肝癌细胞株（Ｑ３）和人膀胱癌细胞株（Ｔ２４）生长的影响。以小鼠成纤维母细胞（３Ｔ３）为正常对照细胞株。实验分为：１．β－胡萝卜素组：剂量为３、３０、１００μｍｏｌ／Ｌ，溶剂对照为ＤＭＳＯ；２．亚硒酸钠组：剂量为０．１５、１．５、５．０μｍｏｌ／Ｌ，溶剂对照为生理盐水；３．β－胡萝卜素和亚硒酸钠联合组：剂量为１．５＋０．１５、１５＋１．５、５０＋５．０μｍｏｌ／Ｌ，溶剂对照为ＤＭＳＯ和生理盐水。各实验组均设空白组。分别在实验后２４ｈ、４８ｈ、７２ｈ观察细胞成活率和活细胞蛋白质含量的变化。结果：１．β－胡萝卜素各剂量组对两株肿瘤细胞生长均有明显的抑制作用；２．亚硒酸钠对两株肿瘤细胞的抑制作用较弱，但随作用时间延长和剂量的增加其抑制作用明显增强；３．β－胡萝卜素和亚硒酸钠对Ｑ３细胞株的抑制作用比Ｔ２４细胞株强；４．β－胡萝卜素和亚硒酸钠联合对Ｑ３和Ｔ２４细胞的抑制作用结果提示两者有相加效应。The　effects　of　βcarotene　and/or　inorganic　selenium(Na2SeO3)on　human　hepatic　carcinoma　cell　line　(Q3)and　human　bladder　carcinoma　cell　line(T24)were　studied　with　technique　of　cell　culture.Fibroblast　cell　line　of　mouse　served　as　normal　control　cell　line.There　were　three　parts　in　the　test:1,Studying　the　effect　of　βcarotene　with　dose　of　3,30,100　μ　mol/L,dissolvent　control　being　DMSO;2.Investigating　the　effect　of　sodium　selenite　(Na2SeO3)with　dose　of　0.3,3.0,10　μ　mol/L,dissolvent　control　being　saline;3.Examining　the　effect　of　the　combination　of　βcarotene　and　sodium　selenite,their　concentrations　being　1.5+0.15,15+15,50+5μ　mol/L,　dissolvent　control　being　DMSO　and　saline.A　blank　control　was　arranged　in　each　group.The　survival　rate　and　the　protein　content　of　the　cultured　cells　were　observed　at　24h,48h　and　72h.The　results　showed　that:1.There　was　a　significant　growth　inhibition　on　both　of　the　two　carcinoma　cell　lines　by　each　dose　group　of　βcarotene;2.The　growth　inhibition　on　cultured　cells　by　inorganic　selenium　was　weaker,but　the　inhibitory　effect　was　significant　by　increasing　dose　and　prolonging　the　cultured　time.The　growth　inhibitory　effect　of　βcarotene　and　sodium　selenite　was　stronger　on　(Q3)　line　than　(T24)　line.The　inhibitroy　effects　of　the　combination　of　βcarotene　and　sodium　selenite　on　Q3　and　T24　carcinoma　cell　lines　indicated　that　there　was　additive　effect　between　them.
