摘 要:研究中国蜂胶醇提物的化学组成及其所含的7种多酚类化合物对酪氨酸酶活性的影响。方法:蜂胶用50%的乙醇提取;用AlCl_3与蜂胶中黄酮类成分的反应产物在415 nm的吸光度检测总黄酮;采用FeCl_3-K_3(FeCN_6)测定蜂胶中的总酚酸;采用RP-HPLC测定蜂胶中有代表性的单体黄酮和酚酸;酪氨酸酶的酶活力测定采用酪氨酸酶多巴速率氧化法。结果:蜂胶醇提物中含有14.1%黄酮和26.6%的酚酸,其有代表性的成分是槲皮素、高良姜素、白杨素、反式阿魏酸、4′,5,7-三羟基黄烷酮、p-香豆酸和反式-4-羟-3-甲氧桂皮酸。当终浓度为0.01 mg/mL时,蜂胶醇提物、p-香豆酸、槲皮素和高良姜素对酪氨酸酶的抑制作用显著,其中p-香豆酸的抑制率明显高于常用的酪氨酸酶抑制剂——熊果苷。白杨素、反式阿魏酸和反式-4-羟-3-甲氧桂皮酸对酪氨酸酶的活性没有显著影响;4′,5,7-三羟基黄烷酮对酪氨酸酶的活性有促进作用。结论:蜂胶含有影响酪氨酸酶活性的有效成分。To investigate the influence on tyrosinase activity of the ethanolie extract of proloplis and its 7 phenolic compounds. Method:Proplis was extracted by 50% ethanol.The flavonoid content was determined by using AlCl_3 to form a complex,which was mea- sured spectrophotometrically at 415nm.The content total phenolic acids was determined by using the FeCl_3-K_3(FeCN_6) regent. RP-HPLC was used to determine the representative flavonoids and phenolic acids.Tyrosinase activity was evaluated by measuring the oxidation rate of L-Dopa.Results:The ethanolic extract of propolis contained 14.1% flavoloids and 26.6% phenolic acids,among which quercetin,galangin,chrysin,ferulic acid,4',5,7-trihydroxy- flavanone,p--coumaric acid and trans--4-hydroxy-3-methoxy cin- namic acid were representative compounds.At the final concentra- tion of 0.01 mg/mL,ethanolic extract of proloplis,p-coumarie acid, quercetin and galangin demonstrated pronounced inhibitory effected on tryrosinase activities,especially p-coumaric acid showed stronger effect than arbutin which is usually used as tyrosinase inhibitor. Chrysin,ferulic acid,and trans-4-hydroxy-3-methoxy cinnamic acid did not lead to inhibition of the enzyme.4',5,7-Trihydroxy-flavanone boosted the activities of tyrosinase.Conclusion:Propolis contained compounds being able to effectively affect tyrosinase activity.
