摘 要:采用PEG/磷酸盐双水相系统从洋葱假单胞菌G-63发酵粗酶液中提取脂肪酶,研究了体系中PEG平均分子量、PEG、磷酸盐、NaCl溶液浓度以及pH对脂肪酶分配系数、回收率、纯化效率及分相时间的影响。结果表明:在PEG2000、磷酸盐、NaCl质量分数分别为10%、15%、1%,pH为8.0时,室温下的双水相体系分配系数为4.36,纯化因子为3.98,脂肪酶的最高回收率达到87.25%。The lipase fermented by P.cepacia G-63 was extracted and purified by PEG/phosphate double aqueous phase system.The effects of the relative molecular mass,concentration of PEG,phosphate,NaCl and pH value on the distribution coefficient,yield,purification factor and phase separation time were examined.The results showed that lipase was recovered with the highest yield when concentrations with mass fraction of PEG2000,phosphate and NaCl in the system were at 10%,15%,1%,respectively,and pH at 8.0.Under the above conditions,the distribution coefficient,purification factor,and lipase yield of the lipase were at 4.36,3.98,and 87.25%,respectively.
