摘  要:为了能够在缺乏Monacolin化合物标准样品的情况下对功能红曲中Monacolin类化合物进行定性、定量分析,建立了用RP-HPLC-PDA检测器测定功能红曲中Monacolin类化合物总含量的检测方法。用75%乙醇超声提取功能红曲中的Monacolin类化合物,高效液相色谱仪以甲醇∶水∶磷酸=385∶115∶0.14(v∶v∶v)为流动相,用反相C18柱(4.6×250mm,5μm)分离提取液中的Monacolin类化合物,并用二极管阵列检测器检测。在此实验条件下,功能红曲提取液中各Monacolin类化合物能够得到良好地分离。用检测器同时给出的各Monacolin类化合物紫外吸收光谱图定性,用外标法根据各Monacolin类化合物峰面积定量。对方法的准确度、精密度、线性关系进行考察;对5种功能红曲样品进行检测均取得良好结果。In order to detect the Manacolin compounds in functional red kojic rice without Monacolin standard qualitatively and quantitatively, a method for detection of Monacolin compounds by RP-HPLC-PDA detector was established. Monacolin compounds in functional red kojic rice were ultrasonic extracted by 75% ethanol. The extraction of Manacolin sample was treated by the RP-HPLC C18 column (4.6×250mm,5μm) for further separation of Monacolin compounds. The motive phase contained methanol, water and phosphate whose mixing ratio was 385:115:0.14 (v/v/v). The separated Monacolin compounds were measured by Photodiode Array (PDA) detector. Under above conditions, different Monacolin compounds in red kojic rice extraction were successful)y separated. The Monacolin compounds were qualitatively detected according to the peak time on ultraviolet action spectrum and were quantitatively detected by the peak area using external standard method. The method was validated with precision, recovery, linearity and accuracy. Five kinds of functional red kojic rice samples were detected by the above method and all gave good results.
