Thapsigargin防治兔眼后发性白内障的研究

摘  要:目的探讨thapsigargin(TG)对兔眼后发性白内障形成的影响。方法所有兔眼均行晶状体囊外摘出术。术后实验组(右眼)晶状体囊袋内分别注入0.1mLTG(A组:0.3μmol/L;B组:1μmol/L);对侧眼(左眼)注入含相同溶剂的BSS液。术后第1、3、7、10、15、35d应用裂隙灯显微镜观察眼前节的炎症反应及晶状体后囊膜混浊情况。术后第35d,Odrich法评估中央区后囊膜混浊程度。摘除眼球行组织病理学检查。结果1μmol/LTG组兔眼后发性白内障的形成明显受到抑制,但存在明显的术后早期眼前节炎症反应(结膜充血、角膜水肿、前房混浊等),于术后第15d左右消退。0.3μmol/L TG组眼炎症反应相对较轻,形成后发性白内障的时间较短,与对照组比较差异无统计学意义(P<0.05)。组织病理学检查结果显示所有兔眼均未发现明显的炎性细胞浸润,1μmol/L TG组兔眼后囊膜区存在增生的晶状体上皮细胞(LECs)。结论高浓度TG能够延缓兔眼后发性白内障的发展,但早期炎症反应较重,毒性反应明显。Objective To find a type of more enduable and effective drug to prevent posterior capsule opacification after cataract surgery is always a hot topic of ophthalmic studies. Thapsigargin was previously proved that it could induce len epithelial cells apoptosis in vitro. The aim of this study was to determine the effect of thapsigargin on development of posterior capsule opacification(PCO) in rabbit eyes. Methods Extracapsular lens extraction was performed on 40 eyes from 20 New Zealand white rabbits. Experimental rabbits were divided into thapsigargin group and balance salt solusion (BSS) group. 0.3 μmol/L thapsigargin was injected into the capsular bag of lens during the operation in 10 right eyes of group A, and the 1 μmol/L of thapsigargin was used in 10 right eyes of group B. The same volume of BBS with equivalent solvent was injected in the same way in the fellow eyes. The anterior segments and the capsule transparency were observed under the slit lamp at day 1,3,7,10,15,35 after surgery, respectively. The intensity of central PCO was graded by Odrich's method 35 days later. The eyeballs were enucleated for histological examination. Results The eonjunctiva congestion, corneal edema and aqueous humour flare were dominant at early stage of operation in 1 μmol/L thapsigargin group in comparison with 0.3 μmol/L thapsigargin group and BSS group(P 〈 0.05 ). Grading of PCO was 1. 385 ± 0. 506 in 1μmol/L thapsigargin group and that in 0. 3 μmol/L thapsigargin group and low or high concentration BSS group was 1. 889 ± 0. 601 , 2.111 ± 0. 782 or 2. 538 ± 0. 660 respectively ( P 〈 0. 05 ). Soemme ring was formed in the periphery of rabbits lens in 35 days. Two layers of proliferation of lens epithelial cells were found under the light microscope in 0.3 μmol/L thapsigargin group and BSS group, and only monolayer proliferation on the posterior capsule was seen in the 1 μmol/L thapsigargin group.
