Notch1与Smad4在大肠癌中的表达及其临床意义

摘　　要：目的:研究Notch1和Smad4的表达与大肠癌临床病理的关系,探讨二者在大肠癌演变中的作用。方法:采用免疫组化方法检测51例大肠癌标本中Notch1和Smad4的表达情况。结果:(1)Notch1的表达明显高于正常组织的表达(P<0.05),大肠癌组织中Smad4的表达明显低于正常组织(P<0.05);(2)Notch1表达与年龄、性别、Dukes分期、淋巴结转移无关(P>0.05),但与肿瘤的大体类型、组织分级有关(P<0.05);(3)Smad4表达与年龄、性别、大体类型无关(P>0.05),但与肿瘤的Dukes分期、淋巴结转移、组织分级有关(P<0.05);(4)Notch1、Smad4联合表达与肿瘤的组织分级有关(P<0.05),与其他临床病理学特性无关(P>0.05)。结论:Notch1的高表达和Smad4的低表达与大肠癌的发展,生物学行为和预后可能有关,可作为重要的生物学标志。Objective： To observe the expression of Notchl and Smad4in colorectal carcinoma and their clinical significance. Methods： Immunohistochemistry staining was used to detect the expression of Notch 1 and Smad4 in 51 cases of colorectal carcinoma. Results： （ 1 ） The expression of Notch 1 in colorectal carcinoma was higher than that in the normal tissue （P〈0.05）.The expression of Smad4 in colorectal carcinoma was lower than that in the normal tissue（P〈0.05 ）. （ 2 ） The expression of Notchl in colorectal carcinoma was not correlated to age and sex of patients, Dukes stage and lymph node of metastasis of colorectal carcinoma （P〉0.05）, except histological grading and pathological types of patients（P〈0.05 ）. （ 3 ） The expression of Smad4 in colorectal carcinoma was not correlated to age and sex of patients, histological grading of colorectal carcinoma（P〉0.05 ）, except Dukes stage, lymph node of metastasis and pathological types of patients （P〈0.05）. （4） The combined check on the expression of Notch 1 and Smad4 in colorectal carcinoma was correlated to histological grading, of patients（P〈0.05）. Conclusion： Notchl and Smad4 may be associated with the carcinogenesis, biological behaviors and prognosis of colorectal cancer. The two factors may be used as important biological markers for colorectal cancer.
