摘  要:目的研究抑癌基因PTEN转染对人膀胱癌细胞系BIU-87化疗敏感性的影响。方法将携有PTEN基因的重组真核表达质粒pBp-PTEN转化大肠杆菌DH5α并扩增,抽提纯化质粒并进行酶切鉴定;pBp-PTEN体外转染BIU-87细胞(pBp-PTEN-BIU87),筛选稳定转染的细胞并扩增培养,以转染了空质粒pBp的BIU-87细胞(pBp-BIU87)和正常BIU-87细胞为对照,用RT-PCR检测PTEN的表达情况,应用MTT法分别研究转染前、后膀胱癌细胞对不同浓度抗癌药物丝裂霉素(MMC)、羟基喜树碱和吡柔比星的敏感性变化。结果RT-PCR证实PTEN在转染的BIU-87细胞中能稳定表达。PTEN转染后,BIU-87细胞对MMC的药物敏感性明显提高,对吡柔比星和羟基喜树碱的药物敏感性无明显变化。结论PTEN基因转染能提高人膀胱癌细胞系BIU-87对抗癌药物MMC的敏感性。Objective To study the effect of transfection with antioncogene PTEN on the chemosensitivity against anticancer drugs of human bladder cancer cell lines BIU-87. Methods The eukaryotic expression plasmid containing PTEN(pBp-PTEN)was introduced into E. coli DHSa and amplified. The plasmid pBp-PTEN was prepared and identified by restriction enzyme. BIU-87 was transfected with pBp-PTEN. The positive cell clones(pBp-PTEN-BIU87) were selected and ampli- fied. BIU87 transfected with empty plasmid (pBp-BIU87) and BIU-87 were employed as control groups,and the expression of PTEN was detected by RT-PCR. Chemosensitivity of BIU-87 against mitomycin C(MMC),hydroxycamptothecin and pirarubicin before and after transfection were measured by MTT assay. Results After transfection with pBp-PTEN,the chemosensitivity of BIU-87 against MMC was increased significantly, but against hydroxycamptotheein and pirarubicin was no change. Conclusions By transfection with PTEN, the chemosensitivity of BIU-87 against MMC was increased.
