摘 要:目的观察β-榄香烯对大鼠肝星状细胞(HSCs)增殖及细胞周期的影响。方法用不同浓度的β-榄香烯对HSCs进行处理,以四甲基偶氮唑蓝法检测细胞增殖,流式细胞仪检测细胞周期,An-nexin-Ⅴ-碘化丙锭染色检测细胞凋亡,逆转录聚合酶链反应(RT-PCR)方法检测HSCs细胞周期蛋白D1(CyclinD1)mRNA、半胱天冬酶2(Caspase2)mRNA的表达。结果2.5、5.0、10.0μmol/L的β-榄香烯均能显著且剂量依赖性的下调CyclinD1mRNA的表达、上调Caspase2mRNA的表达、抑制HSCs的增殖、诱导HSCs凋亡,并可阻滞细胞周期进程,各组G0/G1期细胞百分比随药物浓度逐渐升高。结论β-榄香烯对大鼠HSCs增殖及细胞周期具有调控作用,其机制可能与影响CyclinD1mRNA和Caspase2mRNA的表达有关。Objective To investigate the effect of β-Elemene on the proliferation and cell cycle of rat hepatic stellate cells (HSCs). Methods Rat HSCs were incubated with different concentrations of β-Elemene. Cell proliferation was assessed by MTT colorimetric assay. Cell cycle was analysed by flow cytometry and cell apoptosis was assessed by Annexin-V- propidium iodide. The expressions of Cyclin D1 mRNA and Caspase 2 mRNA were examined by RT-PCR. Results β-Elemene at concentration of 2.5,5.0,10.0μmol/L could down-regulate the expression of Cyclin D1 mRNA, up-regulate the expression of Caspase 2 mRNA,inhibit proliferation, induce apoptosis, inter-fere with cell cycle progression and provoke G0/G1 phase arrest of HSCs in a does-dependent manner. Conclusion β-Elemene can regulate the proliferation and cell cycle of rat HSCs. The mechanism may be partly related to regulating the expression of Cyclin D1 mRNA and Caspase 2 mRNA.
