高效液相色谱法快速测定乳与乳制品中三聚氰胺

摘　　要：目的建立乳与乳制品中三聚氰胺高效液相色谱快速测定方法,为此类检测提供依据。方法乳与乳制品中三聚氰胺经乙腈萃取,采用ZORBAX Eclipse XDB-C8色谱柱分离,以乙腈-庚烷磺酸钠缓冲液作流动相,高效液相色谱二极管阵列检测器（PAD）测定。结果方法线性范围1～50μg/mL,相关系数r〉0.9999,加标回收率为93%～105%,RSD为0.66%,检出限为：奶粉1mg/kg,液态奶制品0.5mg/kg。结论该方法简单、快速,适用于大批量样品的快速检验和乳制品出厂检验。Objective To establish a fast high performance liquid chromatography （HPLC） method for determination of melamine in milk and dairy products. Methods Melamine in milk and dairy products was extracted by acetonitrile, then separated used ZORBAX Eclipse XDB--C8 column （250 ×4.6mm,5μm ） and the mobile phase consisted of acetonitrile - sodium heptanesulfonate, finally analyzed by HPLC with photodiode array detector （ PAD ）. Results The linear range of calibration curves were in rang of 1 -50μg/mL, the correlation coefficient r 〉 0. 9999, the recovery of standard addition were in rang of 93% - 105%, the relative standard deviation （RSD） was 0.66% and the limits of detection were lmg/kg for milk powder and 0. 5mg/kg for liquid milk products, respectively. Conclusion The results showed this method was simpl and rapid and it is adaptable to detect melamine in these products for factory inspection.
