摘　　要：2-氯-6-甲基苯胺与(E)-3-乙氧基丙烯酰氯反应得(E)-N-(2-氯-6-甲基苯基)-3-乙氧基丙烯酰胺,与N-溴代丁二酰亚胺和水、硫脲"一锅法"连续反应制得关键中间体2-氨基-N-(2-氯-6-甲基苯基)-5-噻唑甲酰胺(5)。最后4,6-二氯-2-甲基嘧啶先与5进行亲核取代反应,再与N-羟乙基哌嗪发生亲核取代反应制得抗肿瘤药达沙替尼—水合物,总收率约52%。2-Chloro-6-methylaniline was acetylated with （E）-3-ethoxyacryloyl chloride to give （E）-N-（2- chloro-6-methylphenyl） -3-ethoxyacrylamide, from which the key intermediate 2-amino-N- （2-chloro-6-methylphenyl） -5- thiazolecarboxamide （5） was obtained by one-pot reaction with N-bromosuccinimide and H20, and then thiourea. The chloro groups of 4,6-dichloro-2-methylpyrimidine were successively substituted with amino group of 5, and N-hydroxyethylpiperazine to give antitumor agent dasatinib monohydrate in 52 % overall yield.
