摘 要:制备了交联木聚糖酶聚集体(CLEAs),并对其酶学性质进行了研究,以提高木聚糖酶的稳定性。通过毛栓菌发酵分离得木聚糖酶,经硫酸铵沉淀后以戊二醛作为交联剂对其进行化学交联,制得CLEAs,并用常规方法测定了各种因素对游离木聚糖酶和CLEAs的影响。在对耐热性、有机相中稳定性和耐酸碱性的研究中,CLEAs均表现比游离酶有较高的稳定性,因此运用CLEA技术可提高木聚糖酶的稳定性,使其广泛应用于饲料等工业生产中。In order to enhance the stability of Xylanase, Xylanase was obtained by Trametes trogii's zymosis, then immobilized in a simple and effectiveway by physical aggregation using a precipitant - ammonium sulfate, followed by chemical cross - linking via a bifunctional reagent - glutaraldehyde to form insoluble cross- liked Xylanase aggregates (CLEAs).Tile optimum way of preparing CLEAs was identified, and the enzyme property was studied. Compared with those of flee Xylanase, the thermal stability, stability in organic solvents, and the pH stability, were enhanced. The stability of Xylanase could be highly strengthened by CLEA techniques, it could be used in many industry.
