摘 要:RNA干扰(RNA interference,RNAi)是一种在动植物中广泛存在的通过双链RNA(dsRNA)分子在mRNA水平上诱导特异性序列基因沉默的过程,即转录后基因沉默(post-transcriptional gene silencing,PTGS),由与被抑制基因同源的dsRNA启动。近年来,RNAi作为一种研究工具在生命科学领域得到了广泛的应用,其在医学领域中的应用尤为突出,不仅可应用于基础研究,还有望成为攻克肿瘤、病毒性疾病及遗传性疾病的新手段。现就RNAi应用于妇科肿瘤治疗的研究进展作一概述。RNA interference (RNAi), induced by double-strand RNA homologous to the target gene, is a process of post-transcriptional gene silencing in animals and plants. It plays an important role in down-regulating the expression of the endogenous gene. As a useful technique, RNAi has been widely applied to the study of life science, especially in the field of medicine. It is not only advantageous in basic researches but also a promising technique in fighting tumors, viral infection and inherited diseases. This paper summarizes the mechanisms and characteristics of RNAi as well as the latest progress on its application in gene therapy of gynecological cancer.
