摘  要:为给高抗性淀粉含量小麦品种的选育和遗传研究提供依据,选择抗性淀粉含量差异显著的两个春小麦品种作为杂交亲本构建遗传群体,利用BSA法找到1个与高抗性淀粉含量密切相关的SSR标记Xbarc 59。利用Xbarc 59从F2群体中筛选出高抗性淀粉含量、杂合及低抗性淀粉含量三种基因型,且不同基因型间抗性淀粉含量差异达1%显著水平。根据Xbarc 59引物标记带型分组的高、低抗性淀粉含量的平均值分别为1.44%、0.99%,二者差异达极显著水平,证明Xbarc 59可作为筛选小麦育种材料高抗性淀粉含量的分子标记。Resistant starch(RS) is the sum of starch not absorbed by amylase in the small intestine. It's the focus of carbohydrate researches recently, and the unique physical function of RS has aroused many researchers' concern. The development and exploitation of RS molecule marker technology will reduce the cost and workload of RS determination, overcome the blindness and raise the efficiency of selection, provide theoretical basis to the breeding of wheat varieties with high RS content, and build the foundation of molecular marker-assisted selection of these varieties. In our experiment, M344 and wuchun3 whose RS content were significantly different were used as parents 59 conferring high resistant starch content was screened to develop F2 generations. A SSR marker Xbarc by bulked segregant analysis (BSA) method. In F2 generation of M344/wuchun 3, SSR primer Xbarc 59 is closely correlated to RS content, and the amounts of RS is significantly different at 1% lever among the three genotypes of high-parent type, heterozygote and low-parent type. The average content of higher and lower RS individuals differentiated by Xbarc 59 was 1.44%5and 0.99%, respectively, and the difference is significant at 0.01 level. So the primer Xbarc 59 could be an available molecule marker to select the wheat breeding materials with high RS content.
