摘  要:目的研究Survivin基因在儿童急性白血病细胞中的表达,Survivin反义寡核苷酸对儿童急性白血病细胞凋亡的影响,并探讨反义寡核苷酸与传统化疗药物对白血病的协同治疗作用。方法用PCR的方法检查儿童白血病细胞Sur-vivin表达,将Survivin基因反义寡核苷酸转染入白血病细胞,用TUNEL及流式细胞仪检查各组细胞的凋亡率。结果Sur-vivin基因在儿童急性白血病细胞中高表达,Survivin反义寡核苷酸及足叶乙甙均能够单独诱导白血病细胞凋亡,两者合用时细胞凋亡率更高。结论Survivin在儿童急性白血病细胞中高表达;Survivin反义寡核苷酸可诱导白血病细胞凋亡;Sur-vivin反义寡核苷酸可促进足叶乙甙诱导的儿童急性白血病细胞凋亡。Objective : To research the expression of survivin gene in chilhood acute leukemic cells, the effect of survivin antisense oligonucleotide on chilhood acute leukemic cells' apoptosis, and to explore the cooperated therapeutic action of antisense oligonucleotide combines with traditional chemotherapy drugs for leukemia. Methods: Investigating the expression of survivin gene in chidhood acute leukemic cells by PCR, transfering survivin antisense oligonueleotide gene to leukemic cells, then checking the cells' apoptosis by TUNEL and flow cytometry. Results: The survivin gene has a high expression in chilhood acute leukemic cells, both survivin antisense oligonucleotide and etoposide are able to induce apoptosis respectively, but the leukemic cells have a higher rate of apoptosis when the two are used together. Conclusion: The survivin gene has a high expression in chilbood acute leukemic cells; survivin antisense oligonucleotide can induce apeptosis; Survivin antisense oligonucleotide can promote etoposide - induced apeptosis in in chilhood acute leukemic cells.
