摘 要:目的:探讨超声定位辐照载Fcy::Fur自杀基因微泡对裸鼠卵巢癌皮下移植瘤的抑瘤效应。方法:将荷瘤鼠随机分成5组,超声辐照载基因微泡组、超声辐照基因组、超声辐照微泡组、载基因微泡组和对照组。处理后剖取瘤块测体积、重量,计算抑瘤率;TUNEL检测肿瘤细胞凋亡。结果:各组肿瘤体积、重量、抑瘤率及凋亡均不同,超声辐照载基因微泡组的肿瘤体积、重量最小,抑瘤率最大,凋亡率最高,与其他组比较有显著性差异(P<0.05)。结论:载自杀基因微泡在超声定位辐照下,抑瘤效果好,有望成为卵巢癌新的治疗模式。Objective: To explore the anti - tumor effect of Ultrasound - mediated Fcy: : Fur-loading mierobubble destruction on transplanted ovarian cancer. Methods: 25 mice were assigned randomly to 5 groups, each group received different treatment, ultrasound targeted destrory the geneloaded microbubble group; ultrasoud irradiated gone group;ultrasound irradiated void microbubble group;Gene- loaded mierobubble group ;Control group. After treatment, all the mice were sacrificed and the tumors were harvested to measure volmes, masses and calculate the tumor inhibition rate. Apoptotic cells were detected by TUNEL. Results: The mean tumor volume and mass was the lowest in the group of ultrasound - mediated Fcy: : Fur- loading microbubble destruction, while its tumor inhibition rate and cell apoptosis rate was the highest (P〈0.05 ). Conclusions: Ultrasound - mediated Fcy: : Fur-loading microbubble destruction is an effective therapeutic mode for ovarian cancer. The technique is possible to be adopted as a novel anti - cancer tool for ovarian cancer.
