中国汉族人群蚕蚀性角膜溃疡与HLA-Cw等位基因的相关性研究

摘　　要：目的探讨中国汉族人群蚕蚀性角膜溃疡与人类白细胞抗原(HLA)-Cw基因的相关性。方法收集17例中国汉族蚕蚀性角膜溃疡患者血样,提取基因组DNA。采用序列特异性引物-聚合酶链反应(PCR-SSP)方法来进行HLA-Cw基因分型。正常对照组来源于已知的研究资料。结果蚕蚀性角膜溃疡组HLA-Cw01、Cw02、Cw03、Cw04、Cw07、Cw08、Cw12、Cw14和Cw15的基因频率分别为20.59%、2.94%、26.47%、5.88%、11.76%、17.65%、5.88%、2.94%和2.94%,与正常对照组相比,差异均无统计学意义(P>0.05)。结论在中国汉族人群中,HLA-Cw与蚕蚀性角膜溃疡致病无相关性。Objective Our previous study verified the close correlation of human leucocyte antigens （HLA）-DR17 （3） , HLA-DQ2,HLA-A33, HLA-B58, HLA-DQ0303 and Mooren＇ s ulcer in Chinese Han nationality. However, there still is no the study on whether HLA-Cw is associated with Mooren＇ s ulcer or not. This study was to investigate the association between Mooren＇ s ulcer and HLA-Cw alleles in Chinese Han nationality population. Methods A total of 17 patients with Mooren＇ s ulcer from the same ethnic nationality （ Han nationality） in different parts of China. The diagnosis of Mooren ＇ s ulcer was determined by the typical ulcer manifestation, detail relevant disease history, ocular examination, and outcome of comprehensive laboratory tests. HLA-Cw was typed by polymerase chain reaction（PCR） using sequence specific primers （PCR-SSP）. The HLA typing result was compared with published data from control populations of the same ethnic nationality. Results The gene frequency of HLA-Cw-01,Cw-02,Cw-03,Cw-04,Cw-07,Cw-08,Cw-12,Cw-14 and Cw-15 in Mooren＇ s ulcer patients was 20. 59% ,2. 94% , 26. 47% , 5.88% , 11.76% , 17.65% , 5.88% , 2.94% and 2.94% , respectively. Compared with control population,there was no statistically significant difference between these two groups population. Conclusion Our data suggested that HLA-Cw is probably not associated with Mooren＇s ulcer in Chinese Han population.
