摘  要:目的评价四甲基吡嗪对肝纤维化的防治作用。方法以大白鼠为实验对象,采用60%四氯化碳皮下注射,按每100g体重0.3ml,每3日1次,共23次,诱导肝纤维化模型。四甲基吡预防组和治疗组大鼠分别于第1次、第10次注射四氯化碳时开始给四甲基吡,按每100g体重10mg灌胃,1日1次。实验第10周抽血检测血清中PC-Ⅲ、HA、LN含量,取肝组织作肝内胶原定量分析。结果四甲基吡嗪预防组PC-Ⅲ、HA、LN及肝内胶含量与正常对照组比较。差异无显著性(P>0.05)。四甲基吡治疗组上述四项指标虽高于正常对照组。但明显低于葡萄糖组和肝纤维化模型组(P<0.01)。结论四甲基吡嗪对实验性肝纤维化有防治作用。Objective The purpose of this study was to investigate whether tetramethylpyrazine (TMP) can prevent and delay the development of liver fibrosis in rats. Method Hepatofibrosis model was induced by injecting 60% CCl4 3ml/100g body weight subscutaneously once very three days for ten weeks. Rats in TMP prevention group were given TMP (10mg/100g/day) at the first injection of CCl4. Rats in TMP treatment group were given TMP (10mg/100g/day) at the tenth injection of CCl4. All rats were killed at the end of the tenth weeks. The level of serum procollagen type-Ⅲ(PC-Ⅲ), hyaluronate acid (HA), laminin(LN), the contents of liver collagen were detected respectively . Results No significant difference was noted in the concentration of PC- Ⅲ , HA, LN and the content of liver collagen between the TMP prevention group and the control group (P>0.05). Although the concentration of PC-Ⅲ ,HA,LN and the level of liver collagen were higher in TMP treatment group than those in the control group (P<0.05), it was much lower in the TMP treatment group than that in the control group and the hepatofibrosis model gruop (P<0.01). Conclusion The TMP can prevent and delay the development of hepatofibrosis induced by CCl4.
