摘 要:以负载型结晶尿素为载体,进行了不同脂肪酸乙酯的吸附柱分离,考查了负载型尿素柱分离α-亚麻酸乙酯过程中原料种类、固定相尿素硅胶比例及硅胶粒度等的影响。结果表明:固定相中尿素与硅胶最佳质量比为2:3,以紫苏油乙酯为原料,60-80目硅胶为柱层层析载体,无水甲醇为负载尿素的结晶溶剂,石油醚为流动相.层析柱规格为2cm×60cm,进样量为7mL,洗脱液流速范围为1~1.5ml/min,可得到纯度85.55%的α-亚麻酸乙酯,收率81%。The selective adsorption of fatty acid ethyl ester with different saturation on crystal-urea was studied. Based on this selective adsorption, the feasibility of separation of ethyl α-linolenate by urea column chromatography was investigated, and the factors influencing ethyl α-linolenate separation by urea column chromatography were also studied.The results showed that when the ratio of urea to silica gel in the stationary phase was 2:3 (w/w) ,the raw material was ethyl ester of perilla oil, the carriers was silica gel (60-80 mesh), the load solvent of urea was methanol,the mobile phase was petroleum ether,the column chromatography was 2cm × 60cm,the loading amount was 7mL and the flow rate of eluate range was 1- 1.5mL/min, the purity of ethyl α-linolenate can be up to 85.55% ,and the yield can be up to 81%.
