摘 要:目的观察骨髓间充质干细胞(BMSC)移植对脑缺血后室管膜下区神经干细胞增殖的影响及Notch1表达的变化。方法用线栓法复制大鼠脑缺血再灌注动物模型,随机分为假手术对照组、单纯脑缺血损伤组和脑缺血后BMSC移植组。采用免疫荧光细胞染色法检测各组缺血侧室管膜下区Nestin、Ki-67及Notch1的表达。结果(1)BMSC移植组缺血侧室管膜下区Nestin和Ki-67阳性细胞数较单纯脑缺血损伤组和假手术对照组明显增高;(2)BMSC移植组缺血侧室管膜下区Notch1的表达明显高于单纯缺血组和假手术对照组。结论BMSC移植可促进脑缺血后室管膜下区神经干细胞增殖,且可能与促进Notch1表达有关。Objective To observe the effects of bone marrow mesenchymal stem cells (BMSC) transplantation on the proliferation of neural stem cell (NSC) and Notchl expression in subventricular zone (SVZ). Methods Middle cerebral artery occlusion models were made by intraluminal thread method. The mice were divided into three groups randomly (cerebral ischemia group, BMSC transplantation group and sham operation control group). Immunofluorescence method as used to investigate the expressions of Nestin, Ki-67 and Notchl in SVZ of ischemic hemspnere. Results (1) The proliferation of endogenous NSC in the injured SVZ of the ischemic hemisphere treated with BMSC were significantly higher than those of the cerebral ischemia group and sham operation control group; (2) The expression of Notchl in the SVZ of ischemic hemisphere treated with BMSC were significantly higher than those of cerebral ischemia group and sham operation control group. Conclusion These results suggest that BMSC transplantation can improve the proliferation of endogenous NSC in the SVZ of isehemic hemisphere by promoting the expression of Notchl.
