摘  要:以高脱乙酰度壳聚糖为原料,在乙酸水溶液-乙醇-吡啶介质中,实现了壳聚糖的N位均相乙酰化反应,制备了具有良好水溶性的壳聚糖.研究了乙酰酐用量、反应时间、反应温度、溶剂对壳聚糖脱乙酰度的影响.结果表明,在乙酰酐用量与壳聚糖的摩尔比为2.0、温度40℃、反应时间3 h、溶剂为乙醇、搅拌速度适中时产物的脱乙酰度为52.9%.同时对水溶性壳聚糖进行了红外光谱测定,结果显示52.9%脱乙酰度的壳聚糖与90%脱乙酰度的壳聚糖在分子结构上没有什么不同,52.9%脱乙酰度壳聚糖水溶性的提高是由于部分脱乙酰化对晶体的破坏作用造成的.Water-soluble chitosan with good water solubility was prepared from chitosans with high deacetylation degree in acetic solution-ethanol-pyridine medium by N-position homogeneous acetylation. The effects of acetic anhydride amount, reaction time, reaction temperature and solvent on the deacetylation degree of chitosan were investigated. The result shows that the deacetylation degree of the product is 52.9% under the condition of n(acetic anhydride): n(chitosan) = 2: 1, reaction temperature 40 ℃ , reaction time 3 h, in ethanol solvent and under moderate-speed stirring. The results of infrared spectrometry indicate that there is no difference between water-soluble chitosan with 52.9% deactylation degree and chitosan with 90% deactylation degree in molecular structure; and the improvement of the solubility of the water-soluble chitosan with 52.9% deactylation degree attributes to the destruction to the crystals by the partial deactylation.
