摘 要:杜氏藻(Dunaliella)是生产天然β-胡萝卜素的良好资源,受到世界各国的广泛关注。为了研究和改造其类胡萝卜素代谢途径,利用离心和抗生素筛选相结合的方法对7株生产β-胡萝卜素用的工业藻种进行了分离和纯化。rDNA凝胶电泳图显示,分离的7株藻种中,6株能扩增出0.9 kb^1.7 kb不等的单一条带,表明已分离到单一藻种。在此基础上测定了各藻种转录间隔区1(ITS1)序列,并建立了系统进化树。结果显示,其中3株属于D.viridis类,另3株为D.salina。同时对杜氏藻的生长、工业藻种的分离、D.viridis对生产β-胡萝卜素工业藻的污染及D.salina与D.bardawil分类等问题进行了讨论。Dunaliella is a good resource for natural β - carotene production which attracts worldwide attention. In order to study its carotenoids metabolic pathway and genetic improvement, seven strains of Dunaliella, which produce β- carotene in industry, were isolated and purified combined centrifugation and antibiotic screening, rDNA electrophoresis pattern showed that six of them amplified a single band from 0. 9 kb to 1.7 kb indicating a purified line was isolated. Their Internal Transcribed Spacer 1 (ITS1) was sequenced and a phylogenetic tree based on it was established. The result showed that three of them belong to D. viridis, while the other three belong to D. salina. The growth condition of Dunaliella, the isolation of industrial algal strains, the contamination of D. viridis to industrial algal strains which producting β- carotene and the classification of D. salina and D. bardawil were also discussed.
