Analysis of the Chromosome Constitution of Wheat Lines Resistant to Barley Yellow Dwarf Virus  by Genomic in situ Hybridization

摘  要:利用生物素(biotin-16-dUTP)标记的中间偃麦草(Thinopyrumintermedium)基因组DNA作探针,以未标记的普通小麦中国春基因组DNA作封阻DNA(blockingDNA),对3个抗黄矮病小麦新种质的体细胞染色体进行分子原位杂交。结果表明:抗性源于L1的抗黄矮病小麦新种质Yw642为小片段易位系,含40条小麦染色体和2条小麦-中间偃麦草易位染色体,易位的中间偃麦草染色体片段位于小麦染色体的端部;染色体配对和抗性分析表明该种质为纯合易位系。抗性源于无芒中4的小麦新种质Hw240和Yw060遗传构成不同:Yw060为易位系,含40条小麦染色体和2条小麦-中间偃麦草易位染色体;Hw240为代换易位系,含38条小麦染色体、2条中间偃麦草染色体和2条小麦-中间偃麦草易位染色体。在这2个种质中,易位的中间偃麦草染色体片段均位于小麦染色体端部。The chromosome constitution of three lines (Yw642, Yw060 and Hw240) with BYDV resistance was analyzed by genomic  in situ hybridization (GISH) in which biotin labelled total genomic DNA of Thinopyrum intermedium was used as probe and the unlabelled total DNA of common wheat cv. Chinese Spring as blocking DNA. The results showed that the lines Yw642 and Yw060 were translocation lines. The chromosome composition of these two lines was composed of 1 pair of wheatTh.intermedium  translocation chromosomes and 20 pairs of wheat chromosomes. In both cases, the translocation chromosome segment of Th.intermedium was located at distal end of wheat chromosome arm. The line Hw240 was a substitution /translocation line in which chromosome composition was formed by 38 wheat chromosomes, 2 wheatTh.intermedium translocation chromsomes and 2 Th.intermedium chromosomes.
