摘 要:目的研究不同浓度番茄红素对人宫颈癌HeLa细胞体外增值及侵袭作用的影响,并初步探讨其作用机制。方法不同浓度的番茄红素作用于人宫颈癌HeLa细胞,应用MTT法来检测番茄红素对人宫颈癌HeLa细胞增殖的影响;体外侵袭实验(transwell小室)检测番茄红素对HeLa细胞侵袭转移能力的影响;逆转录聚合酶链反应(RT—PCR)法检测番茄红素对HeLa细胞基质金属蛋白酶-2(MMP-2)基因mRNA表达的影响。结果随着番茄红素浓度的增加,穿过膜的细胞数较空白对照组明显减少,番茄红素能明显降低MMP-2基因mRNA的表达。结论番茄红素对HeLa细胞的生长具有抑制作用,可有效抑制宫颈癌侵袭和转移,其作用机制可能与降低宫颈癌细胞运动能力、抑制MMP-2的表达水平有关。Objective To study the effects of lycopene (Lyp)on proliferation and invasion of HeLa cells and to study the anti - cancer mechanism of lycopene. Methods HeLa cells were treated with lycopene at various concentrations. MTT was used to examin growth and proliferation after treatment with lycopene. Invasion test were used to observe the effects of lycopene on invasion ability of HeLa cells. In addition, the expression of MMP - 2 gene were detected by RT - PCR. Results In the transwell cell culture chambers, invading HeLa cells were significantly lower than those in the con- trol group with lycogen' s concentration. The levels of mRNA of MMP - 2 were decreased. Conclusions Lycopene can inhibit the growth of HeLa cells, lycopene can effectually inhibit the migration and invasion of HeLa cells in vitro. The mechanism may be related to down - regulating the expression of MMP - 2.
