摘  要:应用亲和色谱法、凝胶渗透色谱法及单向免疫扩散法测定牛初乳粉及其产品中免疫球蛋白G质量分数。结果表明,HPAC和HPGPC法的测定结果有良好的相关性,相关系数为分别0.9984和0.9983,稳定性(RSD)为2.01%和3.30%,精密度(RSD)为1.01%和1.45%,重复性(RSD)为1.31%和2.31%,平均回收率分别为97.74%和98.19%。单向免疫扩散法板间变异系数2.11%~4.92%,板内变异系数2.98%~5.69%,平均回收率102.39%。分别用上述方法对样品进行测定,结果显示,凝胶渗透色谱法与亲和色谱法和单向免疫扩散法之间差异均有显著性差异(P<0.05),亲和色谱法和单向免疫扩散法之间差异无显著性差异(P>0.05)。相对而言亲和色谱法具有较快的检测速度,方法简便、准确、样品用量小,可作为控制牛初乳及其相关产品质量的方法。This study applied three methods,a high-performance affinity chromatography (HPAC),a high-performance gel permeation chromatography (HPGPC) and a single radial immunodiffusion (SRID) to determine the contents of IgG in bovine colostrums powder and its products.Results showed that there was a better correlation between the HPAC and HPGPC, correlation coefficients were 0.9984 and 0.9983, stabilities(R.SD) were 2.01% and 3.30%, precisions (R.SD)were 1.01% and 1.45%, repeatabilities (RSD)were 1.31% and 2.31%, average recoveries were 97.74% and 98.19%. The coefficient of variation among plates of SP, ID was 2.11%-4.92% ,whereas coefficient of variation in plate was 2.98%-5.69% and the average recovery rate was 102.39%. The statistical analysis showed that there were significant differences among results obtained from the HPGPC , HPAC and SRID (P〈0.05). However, there was no significant difference between results obtained from the HPAC and SRID (P〉0.05). The HPAC was a method of quick, simple, accurate with litde quantity of sample required. The HPAC was an effective and feasible method determination of the contents of IgG in bovine colostrums powder and its products.
