摘 要:目的:应用血清药理学方法研究千里光抗大肠埃希菌的作用及其机制。方法:从千里光中分离出黄酮类化合物,然后通过给小鼠灌胃制备千里光水浸液的含药血清和黄酮类化合物的含药血清。用扫描电镜观察各含药血清对大肠埃希菌超微结构的影响,用同位素前体参入试验观察各含药血清对大肠埃希菌生物合成的影响。结果:与阴性对照血清相比,2种含药血清作用后的大肠埃希菌,其形态和超微结构均发生明显的变化,表现为菌体表面粗糙,部分菌体出现缢痕,并在缢痕处缢缩或断裂等变化;DNA、RNA、蛋白质和肽聚糖合成也受到明显的抑制。结论:千里光的抗大肠埃希菌作用机制可能是通过抑制细菌的DNA、RNA、蛋白质和肽聚糖的合成有关,其作用的有效成份可能是黄酮类化合物。To explore the antibacterial mechanism of Senecio scandens Buch-Ham(SSB) on Escherichia coil by serum pharmacological method. Method : Flavonoid compound was isolated from SSB, and then the serum contained water decoction agent of SSB or flavonoid compound were prepared with mice. By scanning electron microscope (SEM) , the effects of drug-containing serum on the ultrastrueture of E. coil were observed. By incorporation experiments of isotope-labeled precursor, the effects of drug containing serum on the biosynthesis of E. coil were observed as well. Meanwhile, the serum contained normal saline was set as negative control. Results: Compared with negative control serum, the morphology and uhrastructure of both drug containing serum on E. coil showed obvious changes such as cell surface rough, con- striction or fracture. In addition, the synthesis of DNA, RNA, protein and peptidoglycan was inhibited. Conclusion: The antibacter/al mechanism of SSB on E. coil may be involved with the inhibition of DNA, RNA, protein and peptidoglycan synthesis, its effective ingredients may be flavonoid compound.
