摘  要:盐酸莱克多巴胺(Ractopamine Hydrochloride.RAC)是美国礼莱公司发明的一种新型猪饲料添加剂.具有显著改善动物饲料利用效率.提高胴体瘦肉率的性能.其结构式如图1。猪在上市前喂给RAC.有助于增加胴体重量。提高饲料效率.同时能够保持猪肉的自然口感和嫩汁度。虽然如此.但莱克多巴胺与“瘦肉精”一样危害巨大.我国已明令禁止。Fluorescence immunoassays (FIA) microarrays is a strong candidate platform for antigen analysis because of its ability to measure rare antigens in complex fluids quantitatively. In this paper, agarose film-coated slide was used to detect ractopamine (RAC) in pig urine by indirect competitive fluorescence immunoassay. The glass slide was first activated by the solution with NaIO4 for 30min, then Bovine serum albumin (BSA) conjugates of haptens were immobilized on glass slide. Later, monoclonal antibodies and Cy3 labeled second antibody were added to the glass slide respectively. Finally, the Cy3 generating fluorescence was scanned by chip scanner. According to the Food and Drug Administration (FDA) regulatory guidelines for pharmacokinetic assay validation, this method is quantitative and reproducible with the assay accuracy of 89.09%~111.37%, and its assay precision is less than 20%. The detection limit for analytes was 0.50μg/mL (RAC) , lower than that in the ELISA method.
