摘  要:目的:测定黑柄炭角菌不同组织(天然菌核、天然子座、栽培子座以及发酵菌丝)中的核苷种类及含量。方法:以YWGC18(4.0mm×300mm,10μm)为色谱柱,甲醇-水(甲醇所占体积分数梯度变化为0~8min内由0增加到10%,8~10min内由10%增加到15%)为流动相,流速1ml/min,检测波长259nm,柱温40℃。样品用水超声波提取,0.45μm滤膜过滤。结果:黑柄炭角菌的天然菌核、天然子座、栽培子座以及发酵菌丝中主要含有尿苷和腺苷,子座中的含量高于发酵菌丝和菌核,而且人工栽培的子座与天然子座的核苷含量相差不大。结论:黑柄炭角菌含有丰富的核苷活性成分,并且人工栽培的子座可替代天然子座和菌核。Objective: To determine the kinds and contents of nucleosides in different tissues (natural sclerotium, natural stroma, cultivated stroma and fermented mycelium) of Xylaria nigripes. Methods: Samples were extracted using water in an ultrasonic field, and the extracts were filtrated through 0.45μt m membrane prior to HPLC analysis. The chromatographic separation was achieved on a YWG Cls column (4.0 mm×300mm,10μm) at 40℃ using a mobile phase consisting of a mixture of methanol and water (0 min: 0% → 8 min: 10% → 10 min:15%) at a flow rate of 1 ml/min, and the analytes were detected at 259 nm. Results: The natural sclerotium, natural stroma, cultivated stroma and fermented mycelium ofXylaria nigripes mainly contained uridine and adenosine, and the content of nucleosides in sclerotium was higher than that in stroma and fermented mycelium. Besides, the content of nucleosides in natural stroma was similar to that in cultivated stroma. Conclusion: nucleosides were rich in Xylaria nigripes, and the cultivated stroma can replace the natural stroma and sclerotium.
