摘 要:建立了高效液相色谱法快速测定刺梨中维生素C含量的方法。样品用1g/L偏磷酸超声提取,采用高效液相色谱法,以pH为2·5的0·050mol/L磷酸-甲醇(95∶5)缓冲液作流动相,经C18色谱柱分离,采用紫外检测器,在245nm波长下测定维生素C的吸光值,并与标准系列比较,以保留时间定性,峰面积定量。本方法的线性范围0·050~2000·0μg/mL,检出限为0·005μg/mL,样品加标回收率为88·22%~99·77%,相对标准偏差为0·0135%。本方法具有灵敏度高、准确、快速的优点,5min即可完成定量检测,操作简便,易于普及。To establish a rapid method for the determination of vitamin C in the rosa roxburghii tratt by high performance liquid chromatography.Vitamin C in samples was extracted by ultrasonic with 1g/L metaphosphoric acid solvent.Vitamin C was chromatographically separated on a C18 column with a mobile phase consisting of 0.050mol/L phosphoric acid-methanol(95:5)pH2.5,and then detected at 245nm by ultraviolet-visible detector.The linear range of the proposed method was 0.050-2000.0μg/mL.The detection limit of vitamin C was 0.0051Jg/mL.The average spiked recoveries of the samples were 88.22%-99.77% with the relative standard deviation of 0.0135%.The method is simple, sensitive and applicable to the rapid determination of vitamin C in rosa roxburghfi tratt.The determination of vitamin C in samples could be finished in five minutes.
