摘　　要：应用6-羟多巴胺(6-OHDA)损毁 SD 大鼠单侧黑质制备偏侧帕金森病(PD)鼠模型。3周后根据药物诱发试验,酪氨酸羟化酶(TH)免疫组化证实模型制作成功。并用脑微透析技术结合 HPLC-ECD 在体检测 PD 鼠纹状体多巴胺及代谢产物含量。结果:82只大鼠中有36只阿朴吗啡诱发的旋转次数>7r/min。6-OHDA 注射侧黑质 DA 神经末稍已绝大多数被损毁。6-OHDA 损毁侧纹状体多巴胺及代谢产物明显低于健侧(P<0 05,P<0.01)。结论认为,应用6-OHDA 制备的偏侧 PD 鼠模型是 PD 研究的理想模型之一。Objective:To evaluate the hemiparkinsonian rats induced by 6-OHDA.Methods:Rat models of hemiparkinsonism were induced by unilateral administration of 6-hydroxydopamine(6(?) -OHDA) into the substantia nigra.6-OHDA-treated model rats were successfully proved by apomorphine (APO)-induced rotational asymmetry and TH immunohistochemistry analysis three weeks later.Extracellular dopamine(DA) and its metabolites(DOPAC,HVA) in the striatum of hemiparkinsonian rat model were measured by microdialysis and high performance liquid chromatography with electrochemical detection(HPLC-ECD).Results:36 of the total 82 rats exhibiting the contralateral rotation(>7 turns/min) were selected for further study.Dopaminergic neurons were almost absent in the lesioned nigra and TH-positive fibers were not observed in the denervated striatum.The levels of DA,DOPAC and HVA in the lesioned side were significantly lower (P<0.05,P<0.01,and P<0.01) as compared with those in the intact side.Conclusion:The hemiparkisonian rat induced by 6-OHDA provides a very useful model for the study of human Parkinson disease.
