摘  要:目的构建人乳头状瘤病毒16型(HPV16)E7重组全酿酒酵母疫苗,初步评价疫苗的免疫应答效应。方法将HPV16 E7 cDNA亚克隆入酵母表达载体pYES2/NT,重组质粒转化酿酒酵母,经诱导表达、热灭活制备全酵母HPV16-E7疫苗。疫苗免疫C57BL/6小鼠,ELISA检测其诱导的抗体和细胞因子表达水平。结果重组全酵母疫苗能够有效地表达HPV16-E7蛋白;疫苗小鼠免疫,能够诱导出特异性抗HPV16-E7抗体,并促进Th1型细胞因子的表达。结论研究结果为进一步进行疫苗抗肿瘤免疫研究,提供了实验基础。[Objective] To construct a whole recombinant yeast vaccine with human papillomavirus type 16 (HPV16) E7 gene, and study on the immune response induced by the vaccine initially. [Methods] Subcloned the HPV16 E7 eDNA into yeast expression vector, then transformed recombinant plasmids into the competent Saccharomyces eerevisiae. Positive transformants were induced and heat-inactivated to prepare the whole yeast HPV16-E7 vaccine. C57BL/6 mice were immunized with the recombinant vaccine, the serum concentration of specific anti- HPV16-E7 antibody and cytokines produced from immunized mice was detected by ELISA method. [Results] HPV16 E7 gene could be efficiently 'expressed in the whole recombinant yeast vaccine. The specific anti- HPV16- E7 antibody and much more Thl cytokines from immunized mice could be activated with the recombinant yeast vaccine. [ Conclusions ] This result laid a sound foundation to explore anti-tumor effect of the recombinant vaccine in next step.
