摘 要:针对重组毕赤酵母(Pichia pastoris GS115-phy)表达的基因工程产品的高吨位发酵罐的工业生产,采用称重传感器与计算机组合方式设计并制造了一新型智能称量式补料系统,具有高精度和重复性。智能称量式补料装置与电子秤对比试验误差均在-0.001—0.009kg之间,相对误差小于0.5%。将智能称量式补料装置应用于重组植酸酶发酵,很好地控制了甲醇补料速率,在25m3发酵罐中211h植酸酶表达量达到23,077U/ml的高水平。This paper aimed at high-level expression of recombinant product by yeast Pichia pastoris GS115- phy in industrial-scale fermenter. A novel intelligent fed-batch weighing device was designed and manufactured based on an integration of weighing sensor and personal computer, which is of high precision and reproducibility. For the intelligent fed-batch weighing device, absolute experimental errors ranging from - 0. 001 to - 0.009 kg and relative errors of less than 0. 5% was achieved, respectively, as compared with electronic scale device. Meanwhile, the intelligent fed-batch weighing device was applied to control methanol feeding rate in genetically engineered phytase expression by yeast Pichia pastoris GS115-phy. Recombinant phytase yield of 23077U/ml was reached in 25m3 manufacturing plant at 211h when the intelligent fed-batch weighing device was used.
