纤维蛋白原Bβ-455G/A与新疆维吾尔族、汉族脑梗死的关系

摘　　要：目的探讨新疆地区维吾尔族、汉族健康人和脑梗死患者血浆纤维蛋白原Bβ(FgBβ)-455G/A位点基因多态性频率分布以及与血浆纤维蛋白原水平的关系。方法采用聚合酶链反应限制片段长度多态性(PCR-RFLP)方法对新疆地区156例缺血性卒中患者(维吾尔族63例,汉族93例)和143例的正常人(维吾尔族63例,汉族80例),进行FgBβ-455G/A位点单核苷酸多态性(SNP)分析,磁珠法测定血浆Fg水平。结果(1)新疆维吾尔族、汉族之间FgBβ-455G/A的基因型和等位基因频率分布差异无统计学意义(P>0.05)。(2)维吾尔族、汉族脑梗死组FgBβ-455G/A基因多态性和等位基因频率分布与对照组比较差异有统计学意义(P<0.05)。(3)脑梗死组血浆(fibrinogen,Fg)水平高于对照组,差异有统计学意义(P<0.05),维吾尔族、汉族之间Fg水平无统计学差异(P>0.05)。结论新疆地区维吾尔族和汉族脑梗死患者FgBβ-455G/A基因多态性存在差异,Fgβ启动区-455G/A多态性与脑梗死的发生有关联关系,-455G/A+AA是脑梗死的易感基因型,-455A是易感等位基因。Objective To investigate the frequeney distribution of beta-fibrinogen （beta-Fg） gene -455G/A polymorphisms and their relationship with plasma fibrinogen levels in patients with cerebral infarction in Xinjiang Uygurs and Hans populations. Methods The beta gene Fg Bβ -455G/A polymorphism was identified by polymerase chain-reaction restriction fragment length polymorphism （PCR-RFLP） in 156 patients with CI （including 63 Uygurs and 93 Hans） and 143 health controls （including 63 Uygurs and 80 Hans）. Plasma Fg level was measured by the automatic coagulation analyser. Results （1）There was no statistical difference in the genotype distribution and allele frenquency distribution（P〉0.05） between CI and control group in both the Xinjiang Uygurs and Hans goups.（2） There was statistical significant difference the genotype distribution and allele frenquency distribution （P〈0.05） between CI group and the control group. （3） The plasma Fg level was higher in CI patients with a statistical significance （Uygur： 4.11 ± 0.99g/L vs 3.49 ± 0.97g/L, P〈0.05; Hans： 4.14 ± 1.26g/L vs 3.35 ± 0.87g/L, P〈0.05）. However, there was no statistical significance in the plasma Fg concentration between Uygurs and Hans goups.Conclusion There was statistical significance in the difference of the genotype-455G/A distribution between CI group and the control group, no matter in Uygurs and Hans goups, -455G/A SNPs associate with cerebral infarction course. 455GA＋AA is susceptible genetypes and- 455A is susceptible alleles.
