摘 要:以传统的培养技术分离培养的微生物不到土壤微生物总数的1%,在土壤微生物多样性研究中有很大的局限性。磷脂脂肪酸图谱法、BIOLOG微平板分析法和DNA指纹图谱技术,可描述和表征未被培养的土壤微生物,获得有关土壤微生物多样性的更完整的信息。本文对这三种技术在农田土壤微生物多样性研究中的应用机理、研究现状及其优缺点进行了较系统的概述、分析和探讨。建议在实践中联合使用多种研究方法,综合其优势,确保农田土壤微生物多样性研究方法的有效性和科学性。Less than 1% of microorganisms taxa can be isolated and cultured from soil microbial community by traditional culturing approaches,which had great limitation for studying soil microbial diversity. Phospholipids fatty acid profile analysis,BIOLOG micro-plate analysis and DNA fingerprinting technique,can be employed to describe and characterize soil microorganisms which cannot be cultured currently, and get more perfect information of soil microbial diversity. The application principle,research status,advantage and disadvantage of the three methods were reviewed. Different methods should be combined in order to integrate the advantages of different methods,and ensure the efficiency and scientificity for studying microbial diversity of farmland soil.
