摘  要:目的探讨黄芩甙对重症急性胰腺炎(SAP)大鼠胰腺聚ADP-核糖聚合酶(PARP)活性的抑制作用。方法将45只大鼠随机分为假手术组、模型组、黄芩甙干预组,每组15只。假手术组进腹后仅翻动胰腺和十二指肠后关腹,模型组采用3.5%牛磺胆酸钠逆行胰胆管注射造成SAP大鼠模型,黄芩甙干预组在造模成功后给予黄芩甙治疗。术后3,6,12 h测定各组大鼠血清淀粉酶、脂肪酶及胰腺组织髓过氧化物(MPO)、白细胞介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)的水平,并用免疫组化方法检测胰腺组织PARP的活性代谢产物聚腺苷二磷酸核糖(PAR)的表达水平。结果术后3,6,12 h,模型组血清淀粉酶、脂肪酶及MPO水平均较假手术组显著增高。黄芩甙干预组血清淀粉酶、脂肪酶及MPO水平均显著低于模型组;术后各时间点,假手术组胰腺组织均无PAR表达;模型组PAR阳性,且随时间延长表达显著增多;黄芩甙干预组PAR表达低于模型组,且随时间延长显著减少。术后各时间点,黄芩甙干预组胰腺组织IL-6和TNF-α水平显著低于模型组(P均<0.01)。结论黄芩甙可能通过抑制PARP的活性,减轻活性氧产物对胰腺组织的损害而发挥对胰腺组织的保护作用。Objective It is to explore the inhibition of Baicalin on poly ADP-ribose polymerase(PAPP) in rats with severe acute pancreatitis(SAP).Methods 45 rats were randomly divided into sham operation group(group SO),model group(group M) and Baicalin-treated group(group B),and each group had 15 rats.Rats of group SO were only opened and sutured abdomen without giving any other treatment,rats of the other two groups were conversely injected to biliopancreatic duct with 3.5% sodium taurocholate to made models of SAP. Rats of group B were treated with 5 % baicaline after the models were successfully made. At 3,6 and 12 hours after operation, the serum amylase and lipase were determined, and the con- tents of MPO, IL - 6 and TNF- α in pancreas were determined, and the level of PAR which was the metabolite of PAPP was detected by immunohistochemical examination. Results At 3, 6 and 12 hours after operation, the serum levels of amylase and lipase and the level of MPO in group M were significantly higher than those in group SO, and the levels in group B were significantly lower than those in group M. At different time points after operation, no positive staining for PAR was observed in pancreas tissues from rats in group SO. The expression of PAR in group M was positive, and the expression increased markedly when time went on. The expression of PAR in group B was lower than that in group M, and the expression decreased obviously along with time pass. At different time points, the contents of IL- 6 and TNF-α in pancreatic tissue in group B were significantly lower than those in group M (P 〈 0.01). Conclusion By inhibiting the activity of PARP and reducing the harm that oxygen radical reflects on pancreatic tissue, Baicalin can bring protective effects on pancreas in SAP.
