摘 要:目的:检测乳腺癌及癌旁组织中PTEN基因5’端CpG岛启动子区域的甲基化状况。方法:采用甲基化特异性聚合酶链反应(Methylation-specific PCR,MSP)方法对乳腺癌及癌旁组织中PTEN基因5’端CpG岛启动子区域甲基化状况进行研究。结果:乳腺肿瘤组织中PTEN基因甲基化率为31.5%(29/92),显著高于癌旁组织和正常组织(P<0.05),而癌旁组织和正常乳腺组织中甲基化率均比较低,分别为8.3%(2/24)、6.2%(1/16),两者差异无统计学意义(P>0.05);PTEN基因甲基化与肿瘤组织学类型、年龄及肿瘤大小均无相关性,但与淋巴结转移相关(P<0.05),淋巴结转移组PTEN基因甲基化率显著高于无淋巴结转移组。结论:PTEN基因5’端CpG岛启动子区域甲基化可能是散发性乳腺癌PTEN基因失活的主要机制,参与了乳腺癌的发生发展过程;PTEN基因甲基化可能是乳腺癌发生发展过程中的晚期事件,提示可以考虑将PTEN基因甲基化作为评价乳腺癌肿瘤细胞转移潜能的分子标记。Objective: To investigate the promoter methylation of gene PTEN5 in breast carcinomas and in tissues adjacent to breast cancer. Methods: 92 specimens of breast carcinomas, 24 specimens of tissues adjacent to breast cancer and 16 normal specimens of breast tissues were examined for promoter methylation of PTEN5 with methylation--specific PCR method. Results: Methylation of PTEN was found in 31. 5 % (29/92) of the total breast carcinoma specimens, while in 8.3 % (2/24)of total specimens of tissues adjacent to breast cancer and in 6.2% (1/16) of normal specimens of breast tissues respectively. In addition, promoter methylation of PTEN5 was significantly associated with lymph node metastasis (P〈0.05), but was not associated with differentiation status, tumor size and age. Conclusion: Promoter methylation of PTEN5 is an important mechanism for its inactivation. Methylation of PTEN is a common event in breast carcinomas and may play an important role in the carcinogenesis and progression of breast carcinomas.
