摘 要:[目的]筛选酒精耐受型酿酒酵母菌最佳菌株。[方法]利用紫外线对酿酒酵母JL2008进行诱变处理,并结合氯化三苯四氮唑(TTC)进行筛选,对5株对酒精具有较强耐受性的突变株SC1~SC5的酒精耐受性进行比较,选择出2株酒精耐受性较强的突变株(SC1和SC5)进行酒精发酵,测定其生长速率和发酵性能。[结果]2株菌的酒精耐受能力远远高于出发菌株,均能在酒精体积分数为9%的培养基上生长;在含糖15%的培养基中,SC1的酒精发酵终浓度和发酵效率与原始菌株大致相同;在含糖30%的培养基中,SC1的酒精发酵终浓度和发酵效率分别是16.54%和85.2%,均高于原始菌株。[结论]突变株SC1在含酒精培养基中的生长和在浓醪条件下的发酵效果优于JL2008,是一株极具潜力的应用于浓醪发酵的酵母菌株。[ Objective ] The purpose was to screen out the optimum strain from Saccharomyces cerevisiae with alcohol tolerance. [ Method ] S. cerevisiae JL2008 was mutated by ultraviolet rays and screened out with TFC. Five mutant strains with strong alcohol tolerance (SCI -SC5) were compared. Two mutant strains with the strongest alcohol tolerance( SC1 and SC5 )were selected from these 5 mutant strains for alcohol fermenta- tion,the growth rate and fermentation performance of SC1 and SC5 were determined. [ Result ] The alcohol tolerance of these two strains was much higher than that of the original strain,and they both could grow on the media with 9% ethanol volume fraction. On the media with 15% glu- cose ,the final concn, of alcohol fermentation and fermentation efficiency of SCI were approximately same as that of original strain. On the media with 30% glucose ,the final concn, of alcohol fermentation and fermentation efficiency of SC1 were 16.54% and 85.2% resp. and both of them were higher than that of original strain. [ Conclusion] SC1 was better than JL2008 on the its growth on the media with ethanol and its fermentation effect under the condition of heavy mash, so it was a great potential strain that was applied in the heavy mash fermentation of S. cerevisiae.
