EAU大鼠虹膜睫状体可溶性蛋白的蛋白质吸印免疫学检测研究

摘　　要：目的：观察实验性自身免疫性视网膜葡萄膜炎（ＥＡＵ）大鼠的虹膜睫状体中是否存在视网膜Ｓ抗原。方法：利用视网膜Ｓ抗原特异性的抗血清及免疫偶联辣根过氧化物酶，对虹膜睫状体的可溶性蛋白进行蛋白质吸印免疫学检测。结果：不论正常大鼠，还是ＥＡＵ大鼠，利用本实验所采用的方法，在其虹膜睫状体中，均未发现有与Ｓ抗原特异性抗血清相互反应的可溶性蛋白成分。The soluble proteins in the iris and ciliary body of the normal and EAU rats were analysed with protein immunoblotting (Western Blotting) Various samples of the proteins including the soluble protein in the iris,ciliary body and purified S antigen were transferred from SDS PAGE gels onto nitrocellulose sheets Duplicated samples were treated with anti S antisera,then with HRP IgG The result showed that the purified S antigen reacted specifically with anti S antisera,but no reaction with the soluble proteins in the iris and ciliary body was observed We suggested that the relation between the acute iridocyclitis in EAU and S antigen still need further investigation
