摘  要:为了寻找一种适合于提取马铃薯块茎蛋白质的方法,以常用的5种方法从马铃薯块茎中提取蛋白质,用Brad-ford法测定其中蛋白质的含量,再以SDS-PAGE电泳进行分析,在此基础上对提取过程中所用Tris-HCl提取液浓度进行了优化。结果表明,利用直接提取法所获得的马铃薯块茎蛋白质,不仅含量最高(蛋白质干重为5.54mg),而且电泳后得到的蛋白条带数量最多(达到15条),蛋白条带清晰可见;磷酸缓冲液法次之(4.06mg);酚提取法最差(0.14mg)。此外,以使用浓度为1.5mol/L Tris-HCl的提取液得到的电泳结果最好,条带最多且最清晰。直接提取法提取效率高、操作简便,是适合于提取马铃薯块茎蛋白质的实用方法。In search for a suitable protein extraction method of potato tubers, five common methods for extracting protein from living organisms were used in experiment. The protein content obtained from potato tubers was measured with Bradford method and the character of protein was analyzed by SDS - PAGE. In addition, the concentration of Tris - HCI extraction buffer was optimized for acquiring much better extraction effect. The results indicated that the direct extraction method had the best effective with the highest net weight of 5.54 mg protein, and the most electrophoretie bands (total 15 ) , and all the bands were evidently observed. Phosphate buffer method also showed the better extraction effect with net weight of 4.06 mg protein, however, effect of Hydroxybenzene method was the worst in comparison with other methods which only had net weight of 0.14rag protein. Direct extraction method was an effective, handle -simple, and practicable method for potato tubers protein extraction.
