木糖醇作为渗透泵制剂新辅料的双重作用研究

摘  要:目的:考察木糖醇作为渗透泵制剂新辅料的可行性。方法:考察木糖醇的稳定性及其对药物测定的干扰;分别制备单层渗透泵(EOP)和推拉式渗透泵(PPOP),木糖醇作为促渗剂及致孔剂,并与常用促渗剂和致孔剂比较。结果:木糖醇只在远紫外处有较弱吸收,不易干扰药物的测定;除在强酸和强氧化性条件下以外木糖醇性质稳定;木糖醇的促渗作用与氯化钠相当,调节木糖醇用量可以达到与聚乙二醇(polyethylene glycol,PEG)相同的致孔作用。结论:木糖醇可作为促渗剂及致孔剂用于渗透泵制剂。Objective: To discuss the feasibility of the application of xylitol in osmotic pump preparation. Methods : The stability and the interference of xylitol on the determination of target compound were tested. Elementary osmotic pump(EOP) and push-pull osmotic pump(PPOP) were prepared using xylitol as a penetrating agent and a channel agent. The dissolution behavior of xylitol was compared with the osmotic pump tablets using NaCl as a penetrating agent and PEG as a channel agent. Results:Xylitol was stable except in strong acidic and oxygenic surrounds,which just had a weak UV absorption in short wavelength, thus it did not disturb the measurement of drugs. The penetrating ability of xylitol was as good as NaCl. The ability of xylitol as a channel agent was the same as PEG with an appropriate switch of the amount. Conclusion :Xylitol could be used in osmotic pump preparations as a penetrating agent and a channel agent.
