摘 要:应用rasp21单克隆抗体免疫细胞化学法分析28例急性髓细胞白血病和15例急性淋巴细胞白血病,两者p21阳性率分别为357%和67%,差别有显著性意义(P<005),p21主要在急性粒细胞白血病中表达。在12例慢性粒细胞白血病(CML)中有5例p21阳性,占417%,推测ras基因的扩增可能在CML的发展中起一定作用。免疫细胞化学结果显示:ras基因的损伤并非累及白血病细胞的某一特定阶段或同一发展阶段的全部白血病细胞。ras基因的扩增与白血病细胞的增殖、分化以及白血病患者的预后有关。By applying rasp21 monoclonal antibody immunocytochemical analysis of the ras oncogene protein p21 in leukemic cells,had found p21 positive in 10/28(35.7%) of AML,in 1/15(6.7%) of ALL.The difference of p21 positive rate was singnificant(P<005).The expression of rasp21 was mainly in acute myeloblastic leukemia.Among 12 patients with CML,5 of them were p21 positive(41.7%).It was speculated that the ras oncogene amplification may play some important role in the evolution of CML.Immunocytochemistry displayed that the ras oncogene does not always involve in some one stage leukemic cells or the all leukemic cells that belong to the same developing stage in morpholog.The amplification of ras oncogene may associate with the cell proliferation,differentiation and the prognosis of patients with leukemia.
