施普善对慢性低氧高二氧化碳大鼠学习记忆及海马caspase-3表达的影响

摘  要:目的:观察脑活素(施普善)对慢性低氧高二氧化碳大鼠学习记忆功能的影响及大鼠海马中caspase-3表达的变化,并探讨可能的作用机制。方法:筛选八臂迷宫训练后清洁级的雄性SD大鼠32只,随机均分为对照组、模型组、施普善1 mL/kg组和施普善2.5 mL/kg组。采用八臂迷宫观察干预前后大鼠空间学习记忆能力的变化,Western-blot法检测大鼠海马caspase-3蛋白的表达。结果:与对照组比较,模型组大鼠空间学习记忆能力下降,海马中caspase-3明显增高。与模型组大鼠比较,2种剂量施普善干预组大鼠的学习记忆能力明显改善,且海马中caspase-3的表达减少。结论:施普善具有改善慢性低氧高二氧化碳大鼠空间学习记忆作用,可能与抑制海马中caspase-3表达有关。Objective:To observe the effects of cerebrolysin on the learning-memory function and the changes of caspase-3 expression in the hippocampus of rats model with chronic hypoxia and hypercapnia,and to explore the probable mechanism.Methods: Thirty-two male S-D rats trained by eight-arm radial maze were divided into four groups: rats in the control group without treatment;rats in chronic hypoxia and hypercapnia group were placed in the special closed cabin with hypoxia and hypercapnia for 8 hr a day,6 day a week,4 weeks and inOected intraperitoneally with saline; the rats in two treating group were injected intraperitoneally with 1 mg/kg and 2.5 mg/kg cerebrolysin, respectively. The learning-memory ability was measured using eight-arm radial maze and the expression of caspase-3 in the hippocampus was decteced with Western-blot.Results: Compared with the control group, the learning-memory ability was declined markedly and the expression of caspase-3 was increased significantly in chronic hypoxia and hypercapnia group. Nevertheless, the improvement of learningmemory and the less aaspase-3 expression were observed in rats treated by two doses of cerebrolysin. Conclusion: Cerebrolysin may improve the learning-memory ability in rats with hypoxia and hypercapnia, which may be associated with the inhibition of the exoressin of casease-3 in the hippocamous.
