食管鳞状细胞癌survivin表达与其CpG岛甲基化的研究

摘　　要：目的探讨食管鳞状细胞癌(esophageal squamous cell carcinoma,ESCC)肿瘤组织survivin基因的表达与其CpG岛甲基化状态的关系。方法采用RT-PCR的方法检测ESCC细胞株、ESCC肿瘤组织及对应的远癌组织中的sur-vivin基因mRNA的表达;采用亚硫酸氢钠修饰后测序法(BSP)检测survivin基因CpG岛甲基化状态。结果ESCC肿瘤组织survivin mRNA表达阳性率为65.5%,而绝大部分远癌组织表达阴性;20例survivin表达阳性的肿瘤组织、survivin表达阴性的远癌组织及5株ESCC细胞所扩增片段中的CpG位点均呈非甲基化状态。结论ESCC组织中survivin的表达与否与其CpG岛的甲基化状态无关,提示该区域的甲基化修饰未参与调控survivin基因的转录。Objective To detect the expression of surviviu in the tissues of esophageal squamous cell carcinoma （ESCC） and analyze the methylation status of CpG island in the promoter of survivin. Methods RT-PCR was used to detect the expression of survivin in 5 ESCC cell lines （EC9706, KYSE150, KYSE450, EC109 and TE-1 ） , and 84 samples of paired carcinoma tissues and para-carcinoma tissues. The methylation status of survivin was detected by using bisulfite sequencing PCR （BSP）. Results The results of RT-PCR showed that survivin was expressed in all the 5 ESCC cell lines and 65.5% carcinoma tissues but not in para- carcinoma tissues. However, CpG methylation of the survivin promoter was not detected in all the cell lines, and 20 pairs of carcinoma and para-carcinoma tissues. Conclusion Survivin is overexpressed in ESCC, how- ever this abnormality is not regulated by its promoter methylation.
