摘 要:目的研究平原常氧和模拟高原缺氧条件下慢性牙周炎SD大鼠模型龈沟液(GCF)中可溶性细胞间黏附分子-1(sICAM-1)浓度的变化,以探讨sICAM-1与高原牙周炎的关系。方法选取40只SD大鼠,雌雄各半,分为平原对照组(A组)、平原牙周炎组(B组)、缺氧对照组(C组)、缺氧牙周炎组(D组),每组10只。B、D组大鼠分别建立平原、高原牙周炎模型,A、C组分别作为平原、高原对照组。8周后采用ELISA法检测4组大鼠GCF中sICAM-1浓度变化,并进行比较。结果A、B、C、D组GCF中sICAM-1浓度分别为21.98±2.93、32.20±3.80、23.44±3.81、36.64±2.64ng/ml,B组与A组、D组与C组、C组与A组、D组与B组比较,差异均有统计学意义(P<0.05),其中D组显著高于其他三组(P<0.05)。结论缺氧牙周炎组大鼠GCF中的sICAM-1水平高于平原牙周炎组,提示高原缺氧环境对机体的免疫黏附能力有一定影响。Objective To establish the rat model of periodontitis in condition of plain and simulated plateau condition,and investigate the changes in concentration of soluble intercellular adhesion molecule-1(sICAM-1)in gingival crevicular fluid,so as to explore the relationship between sICAM-1 and plateau periodontitis.Methods A total of 40 SD rats(20 males and 20 females)were randomly divided into 4 groups(10 each):plain control group(group A),plain periodontitis group(group B),plateau control group(group C)and plateau periodontitis group(group D).The models of plain periodontitis and plateau periodontitis were establish according to Fischer's method in groups B and D respectively,and the rats in groups A and C received no corresponding treatment for establish plain control and plateau control models.Eight weeks later,ELISA was performed to determine the changes in sICAM-1 concentrations in gingival crevicular fluid among the 4 groups.Results The sICAM-1 concentration in gingival crevicular fluid was 21.98±2.93ng/ml in group A,32.20±3.80ng/ml in group B,23.44±3.81ng/ml in group C and 36.64±2.64ng/ml in group D,respectively.There were statistical differences between group B and group A,group D and group C,group C and group A,and group D and group B(P〈0.05).The sICAM-1 concentration increased significantly in group D compared with that in the other 3 groups.Conclusion The sICAM-1 level in the rats of plateau periodontitis group is higher than that of plain periodontitis group,implying that hypoxia in plateau areas may create some influence on the immune adherence ability of the body.
