摘  要:目的通过大蒜素作用于人胃癌细胞系MGC-803,探讨其对胃癌细胞增殖的影响及机制。方法通过细胞培养技术检测大蒜素作用胃癌细胞后细胞增殖抑制率;免疫组化及RT-PCR法检测胃癌细胞中p16 INK4因子表达变化。结果大蒜素作用于MGC-803细胞后,对其增殖表现出了明显的抑制作用,且呈浓度依赖性;大蒜素处理的MGC-803细胞p16蛋白及p16 INK4 mRNA表达明显强于未应用大蒜素处理组(P<0.05)。结论大蒜素可以抑制MGC-803细胞的增殖,其机制之一可能为上调了细胞抑癌基因p16 INK4的表达。Objective To study the effect of allitridi on human gastric cancer ceils lines MGC-803 so as to disscuss its effect on the proliferation of human gastric cancer cells and its possible mechanism. Methods To detect the proliferation inhibition of the cells after allitridi affecting gastric cancer cells with technology of cell culture and to examine expressions of the p16INK4 gene using immunohistochemistry and RT-PCR. Results With the effection of allitridi on gastric cancer cells, its proliferation had been resisted by allitridi obviously and in concentration dependent manner. The expression of p16 was more higher compared with the control group. The expression of MGC-803 cell p16INK4 mRNA increased obviously compared with the control group. Conclusions Allitridi could inhibit the proliferation of the cell cycle of human gastric cancer cells lines MGC-803 ,and improve the expression of the anfi-oncogene such as p16INK4.
