摘  要:目的探讨表没食子儿茶素没食子酸酯(EGCG)治疗鼻咽癌的作用机制。方法体外培养鼻咽癌CNE-2细胞并以不同质量浓度EGCG处理,采用MTT法检测细胞增殖抑制率,流式细胞术检测细胞凋亡率,RT-PCR检测细胞中锰超氧化物歧化酶(MnSOD)表达变化。结果EGCG处理后,CNE-2细胞增殖明显受到抑制,且呈时间和剂量依赖性;CNE-2细胞凋亡率显著升高,且呈量效关系;MnSOD表达明显上调,且随EGCG作用时间延长有升高趋势。结论EGCG能抑制CNE-2细胞增殖、促进其凋亡,可能机制为上调MnSOD表达;此为EGCG在鼻咽癌防治中的应用提供了理论依据。Objective To explore the anti-cancer effect and mechanism of (-)-Epigallocateehin-3-Gallate (EGCG) on nasopharyngeal carcinoma. Methods Nasopharyngeal carcinoma cell line CNE-2 were cultured in vitro and treated with various concentrations of EGCG,the proliferation inhibition rate was detected by MTT, the apoptosis was analyzed by flow cytometry,the expression of MnSOD was detected by RT-PCR. Results The CNE-2 proliferation was inhibited in a dose- and time-dependent manner after treated by EGCG, the CNE-2 apoptosis ratio increased significantly in dose-dependent manner. The expression of MnSOD increased significantly in time-dependent manner. Condusions EGCG can suppress the proliferation and induce apoptusis in CNE-2 ,possible mechanism is up-regulating the expression of MnSOD. This study may provide a theoretical basis for the prevention and therapy of nasopharyngeal carcinoma by EGCG.
